RSpondin 3 (RSPO3, roof platespecific spondin 3), also called cysteinerich and single thrombospondin domain containing1 (Cristin 1), is an ~31kD secreted protein that shares ~40% aa identity with the other three RSpondin family members. All are positive modulators of Wnt/betacatenin signaling, but each has a distinct expression pattern. Like other Rspondins, RSpondin 3 contains two adjacent cysteinerich furinlike domains (aa35135) with one potential Nglycosylation site (aa 36), followed by a thrombospondin (TSP1) motif (aa147207) and a region rich in basic residues (aa211269). Only the furinlike domains are needed for betacatenin stabilization. Within aa21209, mouse RSpondin 3 shares 93%, 97%, 96%, 95% and 91% aa identity with human, rat, equine, bovine and canine RSpondin 3, respectively.
