Description
Heparin Cofactor II (HCII) is a plasma glycoprotein that is a member of the SERPIN family of proteinase inhibitors. HCII is a relatively specific inhibitor of thrombin and its activity is increased approximately 1000 fold in the presence of heparin. Its activity is also increased in the presence of dermatan sulfate. HCII is a single chain molecule with a mass of 65.6kD. Inhibition of thrombin results in a covalent complex of 102.6kD. HCII in plasma and other fluids can be measured by capturing onto a microtitre plate coated with an affinity purified antibody to HCII. After washing the plate to remove unbound material, a peroxidase conjugated second antibody is added to the plate which will bind to the captured HCII. After washing of the plate, the peroxidase activity is expressed by incubation with ortho-phenylene diamine (OPD) and after a fixed time the reaction is quenched with the addition of H2SO4.
