Description
Hydrogen/deuterium exchange mass spectrometry (HDX-MS) is an important tool for interpreting the higher order structure and dynamic of biopharmaceuticals. HDX is used to measure the binding interaction between protein and ligand. The presence and absence of ligand in protein sample lead to different extent of isotope exchange, which can be determined in MS analysis. And the binding can be overall studied by change the experimental conditions, like pH, type and concentration of salt, competitive molecules, or amino acid mutation at putative binding sites.
