Matrix metalloproteinase 2 (MMP-2), also known as gelatinase A or 72 kD type IV collagenase, is a member of a broad family of MMPs, which are zinc-dependent enzymes that break down extracellular matrix proteins as part of their primary functions. MMP-2 is secreted in its 72 kD latent pro form from many cell types including neutrophils, monocytes, macrophages, endothelial cells, platelets, and tumor cells. It exists as a monomer and can form a homodimer that is known to regulate pro-MMP- 2 activation. When its pro-domain is cleaved by proteases such as MMP-14 and thrombin, it becomes the active 62 kD mature form. Activated MMP-2 degrades collagenase of the basement membrane and gelatin as well as several chemokines and cytokines. MMP-2 can also bind to other proteins, such as the tissue inhibitors of metalloproteinase (TIMPs). Specifically, TIMP-2 is able to bind to MMP-2 and MMP-14 to form a complex that regulates MMP-2 activation on the cell surface. MMP-2 is closely related to MMP-9, although it only shares 46% amino acid sequence identity. Human MMP-2 shares significant {>95%) amino acid sequence identity with MMP-2 from other species, including non-human primates, equine, porcine, mouse, bovine, and rat. MMP-2 plays important roles in vasculature remodeling, angiogenesis, tissue repair, tumor invasion, inflammation, and atherosclerotic plaque rupture. MMP-2 is a potential biomarker for many diseases and conditions, as it is involved in tumor metastasis, inflammatory conditions, allergies, autoimmune diseases, and myocardial infarction.
