The peptide is synthesized as a 180 amino acid precursor which is converted to glucagon by prohormone convertase 2 (PC2). The primary role of glucagon is in gluconeogenesis and glycogenolysis. It stimulates the release of glucose by converting glycogen in liver, and also maintains glucose homeostasis in coordination with insulin.
