Unit Definition: 1 unit of enzyme is defined as the amount of enzyme that is required to digest 1μg of substrate DNA to completion in 1 hour in a total reaction volume of 50μl, with appropriate assay conditions (salt concentration, pH and temperature). Please note that the activity of the restriction endonuclease is substrate-dependent. The enzyme should be titrated to determine its actual activity when working with a new substrate.
