As an excellent method for evaluation of secondary structure, folding and binding properties of proteins, circular dichroism (CD) spectroscopy measures differences in the absorption of left-handed polarized light versus right-handed polarized light arise due to structural asymmetry. An ordered structure results in a spectrum containing both positive and negative signals, while the absence of regular structure show zero CD intensity.
