Description
Fusion: 1980. Binds to a conformation-dependent epitope on the beta-1 isoform of the Na, K-ATPase. This isoform is expressed in many tissues, including skeletal muscle, heart, kidney, liver, sensory and motor neurons, photoreceptor cells. The epitope is expressed on the cell surface. The antibody can thus be used to label living cells or to quantify sodium pumps in the plasma membrane. The epitope is probably the same as that for mAb 24 (in the DSHB collection from the same lab}. These antibodies compete for binding. mAb 24 has a higher affinity. II-20/5.75.36 is easily iodinated, whereas 24 is very labile under iodination conditions. II-20/5.75.36 cross-reacts with at least some other avian species and fish. Does not cross-react with Na/K pumps of Xenopus, Drosophila, rat, mouse, rabbit, monkey, or human. Does not cross-react with beta-2 isoform. The epitope is conformation dependent and maps near the middle of the 305 aa polypeptide, based upon binding to chimeric human/chick beta subunits. This mAb is not very good for immunoblotting and mAb 24 is superior for immunoprecipitation (precipitates alpha-beta complexes from triton X-100 or C12E8 solubilized cells) or immunohistochemistry.This antibody doesn't seem to affect ATPase function or cell survival.
