MALDI-TOF MS represents a competitive, valuable alternative to ESI-MS because its performance is less affected by buffer components, detergents and contaminants, and allows intact protein mass determination with sufficient accuracy (<500 ppm) for sequence validation. Furthermore, MALDI generates ions containing fewer charges than ESI and therefore acquiring and interpreting MALDI data is more straightforward. The full mass spectrum can be interpreted without any deconvolution step.
